Mounjaro® Tirzepatid

Verursacht Mounjaro® (Tirzepatid) ein medulläres Schilddrüsenkarzinom (MTC) oder eine C-Zell -Hyperplasie?
Es ist nicht bekannt, ob Tirzepatid Schilddrüsen-C-Zell-Tumore, einschließlich MTC, verursacht, da die Relevanz von Tirzepatid-induzierten Nagetier-Schilddrüsen-C-Zell-Tumoren für den Menschen nicht bestimmt wurde.
Wie oft wurden medulläres Schilddrüsenkarzinom und C-Zell-Hyperplasie beobachtet?
Beobachtungen in Nagetierstudien
Eine 2-jährige-Karzinogenitätsstudie wurde mit Tirzepatid an männlichen und weiblichen Ratten in Dosen von 0,15, 0,50 und 1,5 mg/kg (0,12-, 0,36- und 1,02-fache der maximal empfohlenen Humandosis (MRHD) basierend auf der Fläche unter der Kurve) durchgeführt (AUC)) verabreicht durch subkutane Injektion zweimal wöchentlich. Tirzepatid verursachte bei allen Dosierungen im Vergleich zu den Kontrollen eine Zunahme von Schilddrüsen-C-Zell-Tumoren (Adenome und Karzinome).1,2
Beobachtungen in klinischen Studien
Erhöhte Calcitoninspiegel im Blut sind eine bekannte Nebenwirkung von Tirzepatid, die gelegentlich vorkommt (≥ 1/1000 bis < 1/100).2
In den SURPASS-Studien wurden keine klinisch relevanten Veränderungen der mittleren Calcitoninspiegel beobachtet und es wurden keine Fälle von medullärer Schilddrüsenhyperplasie oder Krebs gemeldet.3-9
